It was governed by Tameyori Toki during Sengoku era, and he was an in-law of Yoshitaka Satomi from Awa. As the Satomi clan surrendered at the Battle of Konodai against Ujiyasu Hojo in 1563, Yoshitaka started to support the conqueror Ujiyasu.

Satomi force led by Yoshitaka attacked Mangi Castle, but they failed to take the castle. In 1590, Tameyori supported the Hojo clan when Hideyoshi Toyotomi began invading Odawara Castle.

Hojo clan eventually surrendered after a long siege and handed over Odawara Castle, and Ujimasa and Ujiteru Hojo committed suicide by seppuku. Other clans from Boso who were assisting the Hojo caln ended up being extinguished or their titles got taken away by Hideyoshi Toyotomi and Ieyasu Tokugawa.

Toki clan at Mangi Castle fell by the hands of Tadakatsu Honda, a loyal servant to Ieyasu Tokugawa. Tadakatsu was given the land as a reward for his service and built Otaki Castle there; thus, it led to the abandonment of Mangi Castle.
